SYNTHÈSE ET ÉTUDES DE SONDES OLIGONUCLÉOTIDIQUES DONT LE SIGNAL FLUORESCENT EST MODIFIÉ AU COURS DE L'HYBRIDATION

Abstract : The aim of this work was the synthesis and studies of oligonucleotidic probes linked to fluorescent labels exhibiting modified signals upon hybridization with the complementary sequences. New ODN-cyanine conjugates were synthesized. The influence of different parameters on the fluorescent signal modification was studied: attachment position of the labels to the ODNs (5'- or internucleotidic position), cyanine structure, stereochemistry. A new phosphorylating reagent has been developed and used for the preparation of the 5'-labeled conjugates. Another family of conjugates involving two identical labels at their 5'-ends was obtained. These labels: thiazole orange, pyrene or perylene are possessing intercalating properties. Hybridization of the conjugates with their complementary sequences induces modifications of the interaction between the two labels leading to changes of the fluorescent signals dependent on the label considered.
