Résumé

Acoustic communication is crucial for social interactions. In humans, it has been suggested that abnormal processing of sensory stimuli plays an important role in social behavior of patients suffering from autism spectrum disorders (ASD), which could be explained by altered activity of cortical networks. Among several hypotheses, that of a deregulation of intracellular Ca2+ reserves have been proposed, although these mechanisms remain to be identified. In cells, endolysosomes release Ca2+ through the activation of two-pore channels (TPC) located at their membrane; the role of these channels in the central nervous system (CNS) is largely unknown.Our team is working on a mouse that does not express the TPC (TPC KO). It has very low plasma oxytocin (OT) levels and social behavior problems, especially mothers who show a maternal behavior deficit. We investigated whether these social behavior problems are related to deficits in cortical processing of communication sounds in TPC KO mice and if this deficit worsens during the transition from virgin to mother. For that, I have analyzed the responses of neurons from the primary auditory cortex of WT (virgin and mother) and TPC KO (virgin and mother) mice to mouse vocalizations (pups and adults) and to guinea pig vocalizations, using two techniques: extracellular electrophysiology in anesthetized mice and calcium imaging in awake mice. We observed that the firing rate, the temporal reliability, the response latency, and more importantly the discrimination between mouse vocalizations are altered in these virgin and mother TPC KO mice, compared to WT mice. Some parameters of cortical responses, but not maternal behavior, were rescued by chronic intranasal administration of OT during the first 9 days postpartum in the TPC KO dam. Our results indicate that there are many changes in the processing of communication sounds, including mouse vocalizations, in the auditory cortex in KO TPC mice during virgin-to-mother transition.
La communication acoustique est cruciale pour les interactions sociales. Chez l’Homme, il a été suggéré qu’un traitement anormal des stimuli sensoriels joue un rôle important dans le comportement social des patients atteints de troubles du spectre autistique (TSA), ce qui pourrait s'expliquer par une activité altérée des réseaux corticaux. Parmi plusieurs hypothèses, celle d’un dérèglement des réserves de Ca2+ intracellulaire a été évoquée, bien que ces mécanismes restent à identifier. Dans les cellules, les endolysosomes libèrent du Ca2+ via l'activation des canaux « two-pore channel » (TPC) situés à leur membrane, canaux dont le rôle dans le système nerveux central (SNC) est largement méconnu.Notre équipe travaille sur une souris n’exprimant pas le canal TPC (KO TPC), qui présente un taux plasmatique en ocytocine (OT) très bas et des problèmes de comportements sociaux, notamment les mères qui montrent un déficit de comportement maternel. Nous avons évalué si ces problèmes de comportements sociaux étaient liés à un déficit de traitement cortical des signaux de communication chez la souris KO TPC et si ce déficit s’aggrave lors du passage de vierge à mère. Pour cela, j’ai analysé les réponses des neurones du cortex auditif primaire de souris WT (vierges et mères) et KO TPC (vierges et mères) à des vocalisations de souris (nouveau-nés et adultes) ainsi qu’à des vocalisations de cobayes, en utilisant deux techniques : l’électrophysiologie extracellulaire sur souris anesthésiée et l’imagerie calcique sur souris vigile. Nous avons observé que le taux de décharge, la reproductibilité temporelle, la latence des réponses, et surtout la discrimination entre des vocalisations de souris sont altérés chez ces souris KO TPC vierges et mères, par rapport aux souris WT. Certains paramètres des réponses corticales, mais pas le comportement maternel, ont été restaurés par l’administration intranasale chronique d’OT lors des 9 premiers jours post-partum chez la mère KO TPC. Nos résultats indiquent qu’il y a bien des modifications anormales du traitement des signaux de communication, notamment des vocalisations de souris, dans le cortex auditif chez les souris KO TPC lors du passage vierge à mère.
