Évaluation et essais d'optimisation de l'activité antifongique des extraits d'écorce de Eucalyptus torelliana F. Muell. (Myrtaceae) sur la croissance in vitro de Candida albicans, Candida glabrata, Candida tropicalis

Abstract : Infectious diseases are a serious public health problem in both developing and industrialized nations. Among these infectious diseases are fungal infections (mycosis) which have taken a considerable proportion especially candidiasis caused by yeast of the genus candida. They are most common in human disease and in HIV patients. This necessitates a growing need to find new antimicrobial compounds capable of inhibiting the growth and resistance mechanisms of microorganisms. It is in this context that our research team has tested various plant extracts (aqueous, hydro-ethanolic, hexane, acetate, butanol and chloroform) of Eucalyptus torelliana F.Muell. a Myrtaceae on the in vitro growth of Candida albicans, Candida tropicalis and Candida glabrata. All these extracts showed inhibitory activity against the studied pathogens with high activity in the X1-2 extract (aqueous phase of hexane-water partition). Optimizing X1-2 extract by gel filtration chromatography (Sephadex G50), gave several fractions of which F5 was the most active fraction. For C. albicans MFC = 62,5 μg/mL; IC50 = 06,45 μg/mL. For C. tropicalis MFC = 62,5 μg/mL; IC50 = 02,70 μg/mL. For C. glabrata MFC = 62,5 μg/mL; IC50 = 01,73 μg/mL. GC/MS of the F5 fraction allowed to identify 15 molecules majority of the compounds are terpenes and phenolic compounds including tanins. The preparation method of the extract (X1-2) by partitioning hydro-ethanolic extract in a mixture of hexane-water solvent followed by gel filtration chromatography on Sephadex G50 this method not only concentrate better but also purify and isolate the active principles.
