(. Title and . English, Study of the pathophysiological role of CD8 regulatory T cells in autoimmunity and under

, Most treatments of these diseases are aimed at decreasing immunity. Recently, the therapeutic research has been focused on the activation and expansion of regulatory cells, particularly with the use of interleukin-2 (IL-2) that has been effective for the expansion of CD4 Treg and improvement of clinical signs

, Rational: Other regulatory cell populations have now received attention, and notably CD8 Treg for which evidence that they are involved in AIDs is growing. However, CD8 Treg are less well characterized than CD4 Treg. Objectives: We sought to deeply characterize CD8 + CD25 + FOXP3 + Treg in healthy donors (HD) and under pathological conditions (Rheumatoid Arthritis (RA) and Systemic Lupus Erythematosus (SLE) patients) at steady state and under low-dose IL-2 treatment

, The aim of TRANSREG is the induction of Treg by low-dose IL-2 in AIDs. 10 mL of blood from 4 untreated HD and from 4 RA and 4 SLE patients (recruited in TRANSREG) before and after IL-2 treatment will be collected. CD8 Treg with be fully phenotyped by flow cytometry, purified by sorting and transcriptomic analysis will be performed, Methodology: TRANSREG is a clinical trial performed in Pitié-Salpêtrière hospital
