Changements cellulaires et moléculaires précoces au site d’injection des vaccins : Caractérisation de la réponse innée et adaptative à l’injection d’un MVA recombinant

Abstract : Vaccination is considered as one of the best therapeutic intervention to fight against infectious diseases. However, we still fail to develop protective vaccines for diseases such as AIDS or B hepatitis, which highlights our lack of knowledge of immune processes.For this thesis, we relied on a non-human primate model associated wih a vaccinia virus living attenuated vaccine derived called MVA (Modified Vaccinia virus Ankara). We deciphered innate, but also adaptive immune response using this model.MVA induces an important inflammatory reaction at the injection site and in the blood after intramuscular, subcutaneous, and intradermal administration. However, the magnitude of this immune reaction, as well as cellular and molecular effectorsin varies depending on the administration route. The consequences are important and lead to different adaptive immune profiles, with Th2 oriented response after subcutaneous route, and Th1 oriented response after intramuscular and intradermal route. It allowed us to highlight several cellular and molecular signatures that might modulate immune response in our model.
