L'approche métabolomique par RMN pour la recherche de biomarqueurs : une étude comparative in vitro du Liquide Céphalo-Rachidien et du sérum de patients atteints de Sclérose en Plaques

Abstract : Multiple sclerosis (MS) is a demyelinating disease which affects specially the central nervous system. The MS diagnosis is difficult to confirm, because the clinical aspects are similar to some neurological diseases. To confirm the diagnosis of MS, the patient has to manifest a temporal criteria (an increase in the number of crisis in time) and a spatial criteria (an increase of the number of neurological lesions visible in MRI). A biochemical criterion, in addition to the clinical manifestations, based on the evidence of the immunoglobulin isoelectric focusing, could confirm the diagnosis. The comparative analysis with 1H NMR of sera and CSF performed on 10 healthy patients and 12 MS patients. Wewill conduct amultivariateanalysis ofspectral data, to search fordiscriminatingmetabolitesaccording topathological conditions. An increase in amino-acids such as isoleucine, valine and tyrosine, then in 2-hydroxybutyrate and the decrease of glucose were observed in the serum of MS patients. They could become interesting biomarkers of the disease.
