Expression différentielle de la GPx5, un membre de la famille multigénique des glutathion peroxydases de mammifères

Abstract : Using various molecular approaches, we found that the mouse gpx5 gene gives rise to at least three different transcripts in the mouse epididymis. In addition to the major gpx5 transcript coding the mature GPx5 protein, two minor gpx5 transcripts exist. Furthermore, we demonstrate that variants of the GPx5 protein that are correlated with the shorter gpx5 transcripts are subject to differential post-transcriptional maturation processes in the mouse epididymis that essentially involve the addition of O-glycosyl extensions. We have demonstrated that the gpx5 gene is also expressed in other tissues of the male genital tract, including the testis and prostate. Finally, obtaining various stages of mouse embryos, we present evidence suggesting that the gpx5 gene is expressed during early embryonic development. This embryonic gpx5 expression relates to a 4th variant whose 3' UTR sequence could not be determinate. Other complementary immunohistochemistry analyses are necessary to confirm the GPx5 protein expression in early mouse embryo and its localization in parietal endoderm.
