Etude de la structure des tissus musculaires par polarimétrie de fluorescence

Abstract : Light interactions with biological tissues have been exploited in an increasing number of techniques for characterization. Among these techniques, uorescence polarization measurement offers a selective way for characterizing the organization of biological tissues. The aim of this work was to build a methodology for investigating the uorescence polarization of muscle tissues in order to obtain information on their structural organization and its modi cations caused by processing. A review presents the principle of uorescence anisotropy and the structure and optical properties of muscle tissues. It highlights variety in published studies concerning optical characterization of structured materials. A detailed report on the experimental methodology is made. A theoretical model of uorescence anisotropy is proposed. It is based on geometrical repartition and properties of tryptophan, which is the major uorophore in muscle bres, and makes the distinction between "intrinsic" and "structural" uorescence anisotropies. Data from several experiments demonstrate how the method is able to show muscle structure modi cations : protein denaturation induced by maturation displays changes in the values of the physico-chemical variable of the model, whereas sarcomere length is related to structural variables of the model. Structure modi cations of connective tissue during heat treatment are also shown. Results show evidence of potential applications for this generic methodology proposed for non destructive assessment of organized materials.
