Préparation d'anticorps anti-acide tiénilique chez le lapin et recherche d'anticorps anti-médicaments dans le serum de patients atteints d'hépatite autoimmune secondaire à la prise d'acide tiénilique

Abstract : Tienilic acid (TA) is a diuretie drug responsible for autoimmune hepatitis in a few subjects. The sera of patients contain auto-antibodies called anti-LKM2 wich recognize cytochromes P450's. These P450's also catalyze the metabolic activation of TA. We describe the haptenization of bovine serumalbumin with TA (BSA-AT). This protein injected to rabbits produces the formation of antibodies (anti-TA) which recognize TA metabolites bound to microsomal proteins as shown by Elisa. These antibodies should be an efficient tool to investigate covalent binding of TA. With BSA-AT and by Elisa, we have looked for the possible occurrence of antibodies raised against TA in the sera of patients suffering from hepatitis. Our preliminary results show that all the sera of patients treated with TA recognize BSA-AT and that the control sera do not.
