Etude du mécanisme d'action des facteurs de remodelage de la chromatine par micromanipulation et visualisation de l'ADN

Abstract : We have studied the behaviour of the chromatin remodeling factors RSC and ISWI using single DNA molecule micromanipulation setups (Magnetic Tweezers and TPM) and AFM imaging. Chromatin remodeling factors are enzymes responsible for regulating chromatin accessibility in the cell. In the first part of this thesis, we introduce DNA structure and the DNA compaction inside the cell. Next, we introduce chromatin remodeling, focusing in particular on ATP dependent remodeling. Next, we introduce standard single molecule techniques used to investigate protein/DNA interactions, describing the setups used to conduct experiments. These differents aproaches allow us to well characterize chromatin remodeling factors translocation, and they allow us to propose a possible mechanism of action to explain chromatin remodeling by the SWI/SNF and ISWI families.
