Régulation UV-dépendante des gènes de la pigmentation. Implication du facteur de transcription USF-1 (Upstream Stimulating Factor 1)

Abstract : The skin is the first protection against physical, chemical and biological hazardous from environment. Protection by skin against solar UV, which accounts for major DNA damage of epidermal cells, is usually obtained with the melanin. Skin pigmentation is based on cellular cooperation between melanocytes and neighbor keratinocytes. Molecular mechanisms involve constitutive pigmentation pathways, which determine specific phototype, but also a UV induced pathway. The implication of the stress-responsive p38 kinase pathway and the transcription factor USF-1 in Tyrosinase gene expression, led us to study the role of the transcription factor in pigmentation (Galibert et al., EMBO, 2001). Several experiments (Real time PCR, Chromatine Immuno-Precipitation, Transfections...) were performed in vitro and in vivo (with cell culture or skin biopsies), with further genetic approach by using USF-1 knock-out melanocyte cell line, and allowed us to establish an UV-induced pigmentation model (Corre et al., JBC, 2004 ; Corre et al., soumis). Finally, the identification of a new stress-responsive post-translational modification of USF-1, which is dose and time dependent, permit to consider a new mode of regulation for USF-1 dependent gene expression (Corre et Galibert, PCR, 2005 ; Corre et al, en préparation). Full results obtained during my PHD complete and specify the function of the transcription factor USF-1 in stress response, and mean to suggest a potential role in melanoma.
