La kinase Aurora A comme nouvelle cible des agents chimiothérapeutiques Application aux traitements des cancers colorectaux

Abstract : The mitosis entry is associated with increase of Aurora A kinase expression. In this study, we characterise the effects of sn38, the active metabolite of irinotecan, the main drugs used in the treatment of colorectal cancer. Without treatment, the transcription factor c-myc actives the Aurora A promoter. However, the treatment of cells with sn38 induces inhibition of c-myc and prevents his association with Aurora A promoter. This treatment is also associated with induction of senescence. On the other hand, we study Ras oncogene effects on Aurora A. We observe a decrease of Aurora A expression through modulation of c-myc transcriptional cofactors activity. Our results show that Aurora A is a target of chemotherapeutics treatment and oncogenic pathways. Blocking Aurora A expression is certainly a protection against tumor development.
