Etude comparative des thérapies anti-VIH : rôle des transporteurs d'efflux sur le passage transmembranaire des antirétroviraux au niveau des cellules CD4+ et de la barrière hémato-encéphalique.

Abstract : HIV protease inhibitors (PI) need to penetrate into the cells at concentrations high enough to inhibit viral replication of HIV-1.
Therefore their transport across the cell membrane might limit their antiviral efficacy. The expression of efflux protein on the surface of cell membranes is expected to reduce intracellular concentrations of PI and to favour the existence of some sanctuary sites.
In this work, we described the differences in intracellular pharmacokinetic of PI and the influence of P-glycoprotein on PI intracellular accumulation. We found that PI went through membrane by passive diffusion and accumulated within cells by linking to cytosolic proteins.
We used cell lines and also peripheral blood mononuclear cells from HIV-infected patients treated by highly active antiretroviral therapy including PI.
