Intérêt du détecteur à dichroïsme circulaire en chromatographie liquide et du détecteur conductimétrique à couplage capacitif en électrophorèse capillaire pour l'analyse de molécules chirales. Applications aux composés pharmaceutiques et aux pesticides

Abstract : The resolution of enantiomers is necessary due to considerable differences that may exist between two enantiomers in terms of pharmacokinetics, activity and toxicity. The development of efficient, specific and sensitive analytical methods is required in order to detect some traces of inactive enantiomer during manufactured batch of medicinal products.
The use of circular dichroism detector in liquid chromatography allowed the simultaneous determination of optical and chemical purities of several drugs. The sensitivity is influenced by several factors (nature of analyte chromophore, pH of the mobile phase, temperature of the detection cell, use of electronic filter). Despite a lack of sensitivity, circular dichroism detector allowed a fast analytical development for the determination of enantiomeric purity, without resolving enantiomers on a chiral chromatographic support.
Furthermore, the ability of capillary electrophoresis to separate pesticide enantiomers which possess heteroatoms as chiral centers was confirmed. Hence, enantioselective separation of one organophosphorus pesticide and its two metabolites has been performed. This method was pre-validated and experiments have been carried out on environmental samples.
Finally, the ease of use and the sensitivity of capacitively coupled contactless detector offer some advantages to determine the enantiomeric purity of amines which do not possess any chromophores by capillary electrophoresis.
