Ligation chimique sur support solide : vers la préparation d'analogues de la glycoprotéine MUC1

Abstract : The work described in this manuscript takes place within the framework of news approaches for anti-tumor immunotherapy. It was aimed at developing a methodology to synthesise mimics of the tumor-related glycoprotein MUC1 by chemical multiligation on solid support. The main strategy was based on a convergent approach through successive condensations of an aminooxy (Aoa) peptide on a peptide aldehyde leading to oxime bonds. The orthogonal protection of the Aoa group of the median fragment enabled several MUC& peptide to be successively coupled by chemical ligation on solid support. Two long peptides with two units and a half repetition of the MUC1 protein for one and one epitope T-additional auxiliary for the other compound were obtained.
This work paves the way for the synthesis of MUC1 chimeras glycosylated by chemistry and enzymatic reaction. These small synthetic glycoproteins, although modeled on natural glycoproteins, will be developed in order to study various presentations antigens to optimize their immunogenicity.
