ROLE DU NAADP ET DE SON ENZYME DE SYNTHESE, L'ADP RIBOSYL CYCLASE, DANS LES NEURONES : LA REGULATION DE L'HOMEOSTASIE CALCIQUE NUCLEAIRE.

Abstract : Calcium is a second messenger, implicated in the regulation of many neuronal functions and has a particularly important role in regulating gene expression. In some cases, gene activation requires nucleoplasmic calcium elevation. In cells, calcium rise can results of calcium mobilization from intracellular stores by second messengers like IP3, cADPR or NAADP, which has been more recently discovered. During my PhD, I show that in neurons, Aplysia ADP ribosyl cyclase, the enzyme synthesizing cADPR and NAADP, can translocate to the nucleus following depolarization and calcium entry through L type calcium channels. Moreover, by using confocal microscopy and calcium fluorescent dyes, we observed in isolated neuronal nuclei that second messengers were able to mobilize calcium from the nuclear envelope in order to generate nucléoplasmic calcium oscillations. Our pharmacological data prove that NAADP activates its own receptor which cooperates with RyRs and IP3Rs to elicit these calcium signals. Finally, to explore the role of NAADP in the general neuronal physiology, I developed a new model of mouse embryonic spinal neurons. Our preliminary data enable us to put an assumption on the implication of NAADP in some important neuronal functions regulated by neurotrophins.
