Caractérisation et analyse fonctionnelle de nouveaux gènes cibles du facteur de transcription hStaf/ZNF143

Abstract : The transcription factor Staf and its human ortholog ZNF173 are involved in the transcriptional activation of snRNA genes transcribed by RNA polymerase II and III and of 7 protein genes. The goal of our study was to characterize new target genes of this factor in humans. Combining a bioinformatic screen and ChIP experiments, we showed that at least 10% of human mRNA genes bind hStaf/ZNF143. This suggests that the staf binding site (SBS) constitutes one of the most widespread transcription factor binding sites in mammalian promoters. Moreover, we studied the promoter of the genes TFAM, BUB1B and SCARNA2 in which we identified numerous conserved elements including SBS. We showed the transcriptional importance of these elements. These results show that Staf regulates the transcription of genes involved in various cellular pathways and via promoters harboring various architectures
