Développement d'outils pour l'étude de la dynamique de l'appareil de Golgi en interphase et en mitose

Abstract : The Golgi apparatus is a central organelle in the web of intracellular transport pathways of eukaryotic cells. The study of its complex dynamics requires the development of new tools. I have therefore followed several strategies in parallel to propose new approaches to study the Golgi apparatus.
I have first used RNA interference, a method enabling the specific depletion of a protein of interest, and thus revealed the complexity of Golgi traffic regulation by the GTPase Rab6.
I have then followed a strategy relying of the in vivo imaging of the disassembly and subsequent reassembly of the Golgi apparatus occurring in mitosis, and thus I have outlined a particular step of the disassembly process.
Finally, my thesis work has essentially consisted in producing recombinant antibodies against Golgi proteins by Phage Display and setting up their use to track the behavior of target proteins in vivo.
