ETUDE DES MECANISMES DE REGULATION DE LA SECRETION ET DE L'EXPRESSION DES MUCINES GASTROINTESTINALES PAR LA LEPTINE

Abstract : Mucins are the basic components of the mucus layer which ensures the protection and lubrication of the gastrointestinal mucosa. Our aim was to study the leptin effect on gastrointestinal mucins secretion and expression. This study was carried out in vivo in the rat and in vitro using the DHE and the HT29-MTX cell lines. We found that these cell lines express the leptin receptors. Luminal perfusion of leptin in the colon of rat or stimulation of DHE by leptin increased mucin secretion and the mRNA levels of rMuc2, rMuc3 and rMuc4. This effect of leptin was dose dependent. In the HT29-MTX, leptin produced an increase in mucin secretion in the supernatants as well as MUC2, MUC4 and in MUC5AC mRNA by targeting PKC, PI3K and MAPK pathways. These results showed that leptin could play an essential role in the protection of the colonic mucosa.
