Nouveaux développements pour l'imagerie par spectrométrie de masse MALDI.
Applications aux problèmes biologiques et à la recherche de biomarqueurs dans le cancer de l'ovaire.

Abstract : MALDI imaging mass spectrometry is a young technique with a high potential, able to give the localisation of more than 100 compounds in tissue in one analysis. Applied to the proteomic or biomedical domains, such a technique has already allowed finding several biomarkers regulated in the cases of cancers or other pathologies. Other applications like the study of drugs or lipids have also been described using the Imaging or profiling studies i.e. acquisition directly on the tissue by the MALDI mass spectrometer. Nevertheless, like all recent technologies, it is necessary to perform new development to improve it. The goal of my work was on the choice of the matrix, sample treatment in order to get a better acquisition quality. The possibility of perform examination of tissues fixed and embedbed in paraffin conserved in hospital libraies was also a challenge. We also developed a new concept, call Tag-mass for specific imaging. All these developments were used in ovary cancer study.
