Etude de la protéine IQGAP1 dans un contexte physiologique de la neurogenèse adulte et dans un contexte pathologique de tumeurs cérébrales.

Abstract : Stem/progenitor cells possess a high cellular plasticity which allows them to develop, maintain and regenerate the organ or tissue inside which they reside. These processes require the integration of molecular and environmental signals which impact on their behaviour and their fate. Alteration in one of these regulatory mechanisms leads to a loss of stem/progenitor control and could give rise to the development of cancers. Therefore, a deeper understanding of molecular and cellular elements regulating stem/progenitor cell biology is necessary to use them in regenerative medicine and to progress in cancer treatments. Our studies reveal that, in the brain, in physiological and pathological contexts the protein IQGAP1 is a new relevant marker of normal and tumour stem/progenitor cells. Through a comparative study between wild-type and iqgap1-/- mice, we analysed the in vivo as in vitro neural stem/progenitor cells properties and behaviour. We concluded that IQGAP1 plays a role in adult neurogenesis by regulating VEGF-dependent neural progenitor chemokinesis. On the other hand, in human and in rat-induced malignant glioma IQGAP1 identifies a tumour cell subpopulation with stem-like cell features that are able to reform a tumour. We suggest a putative role for IQGAP1 in tumour expansion through dissemination of these cancer cells. Identification and characterisation of environmental processes which regulate motility and migration of neural precursors could help to improve our understanding of tumour invasion and to develop more effective anti-cancerous therapies.
