Nanocristaux semi-conducteurs et interaction ADN/EcoRV

Abstract : This report brings together the results of my research activity for the last ten years in the Kastler Brossel laboratory. The work describes in this report was essentially carried out within the team named `` Optique and Biologie''. Our work focuses on the detection of single molecules by optical means and applications to biology using new fluorescent probes whose optical properties are remarkable: semiconductor nanocrystals (Quantum Dots or QDots). I concentrated on the visualization of DNA-protein interactions. Our approach consists in using stretched DNA molecules specifically attached by their ends to a surface. Our goal is to visualize directly by fluorescence microscopy the movement along the DNA molecule of a protein labeled with QDots or with organic dyes. The proximity between surface and the DNA enables us to visualize this movement by Total Internal Reflection Fluorescence Microscopy. We have concentrated on the restriction enzyme EcoRV and in particular on the `` facilitated diffusion'' of this enzyme on stretched T7 DNA. We managed to observe the diffusion of EcoRV, labeled with QDots, along the stretched DNA. We showed that the sliding of enzymes along the DNA is the most probable mechanism which underlies the facilitated diffusion of EcoRV. We also observed large jumps of the enzyme along the DNA. The analysis of the distribution of these jumps enabled us to propose a new vision of the facilitated diffusion.
