Etude d'un continuum de lumière en régime femtoseconde. Applications au domaine biologique : microscopies et spectroscopie en temps résolu.

Abstract : For few years, a new type of source, called "continuum of light", has been developed. It presents many advantages like a wideband spectrum, a huge brilliancy, a pulsed nature. The aim of this work is to perform a complete characterization of a continuum generated by the injection of femtosecond pulses into a microstructured fiber, and to consider the use of this continuum as optical source for biomedical applications.
With acousto-optic filters, the continuum has been successfully employed as tunable source for fluorescence microscopy. Confocal images of biological samples, which were marked by several fluorophores, were recorded. Moreover, we studied and checked the use of the continuum in a STED microscopy set-up, allowing the acquisition of ultra-resolved images with a spectral versatility.
A third application is an optical probe, developed to realize time of flight infra-red spectroscopy on in vivo turbid media, like muscle, bone, marrow of bone... This type of instrument allowed the retrieval of physiological parameters characteristic of the energetic metabolism of those tissues.
