Analyse de l'expression d'une nouvelle cytokine, l'interleukine-27,
dans les lymphocytes normaux et tumoraux
et de son rôle au cours de la différenciation lymphocytaire B normale

Abstract : Interleukin (IL)-27, composed of EBV-induced gene 3 (EBI3) and p28, plays an important role in T cell responses regulation. Its role in B cell responses is not known. EBI3 is induced in vitro by the EBV oncogene LMP1 in a nuclear factor (NF)-kappaB-dependent manner. In this study, we show that EBI3 expression is also induced by the HTLV-1 oncogene Tax, in a NF-kappaB- dependent manner and that is expressed in situ by tumoral cells in EBV+LMP1+ lymphomas and in some HTLV-1 associated lymphomas, without detection of P28. We show here that IL-27 has direct effects on human B cells and that the B cell response to IL-27 is modulated during B cell differentiation and varies depending on the mode of B cell activation. EBI3 but not p28 is expressed by germinal center B cells and by germinal center derived B cell lymphomas. These results suggest a role for EBI3 independently of its association to p28.
