Des puces à protéines/peptides pour des applications en recherche fondamentale et clinique

Abstract : Microarrays are promising devices for high throughput analysis, especially, proteins/peptides chip which is a useful tool for biomedical interactions studies. The chip was designed by electrocopolymerisation of pyrrole and pyrrole-modified biomolecules on gold surfaces. The chip was used to analyse protein-protein or antigen-antibody interactions by surface plasmon resonance imaging which is a direct, real time method suitable for parallel analysis. This method allows dynamic studies to follow the association and the dissociation rates from kinetic curves. This work shows the efficiency of such microarray with high specificity, good sensitivity and possibility of regeneration. Moreover, we demonstrated its compatibility with the analysis of complex biological samples such as undiluted serum. Taken together, these characteristics make this method a powerful tool for numerous applications, including fundamental or clinical researches as well as diagnosis.
