Evaluation par PCR de l'activité antivirale des inhibiteurs de l'ADN polymérase du Virus d'Epstein-Barr

Abstract : A system for the in vitro evaluation of the anti-EBV drugs activity was developed using a quantitative real-time PCR/RT-PCR. Three compounds, belonging to different antiviral classes, were tested: a nucleoside analogue, ganciclovir (GCV), a nucleotide analogue, cidofovir (HPMPC), and a pyrophosphate analogue, foscarnet (PFA). After a 7-day treatment of P3HR-1 virion-producing cells, the calculated concentrations inhibiting viral DNA replication by 50 % were 0.28 µg/mL, 0.29 µg/mL and 13.6 µg/mL for GCV, HPMPC and PFA respectively. The expression of the late viral glycoprotein gp350/220 mRNA, was reduced by 79 to 89 % after 4 days. We have also shown the specificity of the antiviral effect by measuring the amount of cellular DNA (b-globin) or RNA (hPBGD).
In conclusion, our system permits the evaluation of the antiviral effect against EBV in a quantitative, simple and accurate manner.
