CARACTERISATION DE LA PROTEINE ALIX ET DE LA MACHINERIE ESCRT CHEZ L'AMIBE DICTYOSTELIUM DISCOIDEUM.
UN LIEN ENTRE ENDOCYTOSE ET SIGNALISATION DEVELOPPEMENTALE?

Abstract : IN THIS STUDY, WE HAVE CHARACTERIZED THE MULTIMODULAR PROTEIN ALIX IN DICTYOSTELIUM DISCOIDEUM. IN THIS EUCARYOTIC ORGANISM, THE DEVELOPMENTAL CYCLE INCLUDES A MULTICELLULAR PHASE THAT INVOLVES BOTH DIFFERENTIATION AND A PROGRAMMED CELL DEATH. THE DISRUPTION OF ALX LEADS TO A MAJOR DEVELOPMENTAL DEFECT AND AN ABERRANT MORPHOGENESIS BUT THE PROGRAMMED CELL DEATH IS NOT IMPAIRED. A STRUCTURE/FUNCTION APPROACH REVEALS THAT THE COILED-COIL DOMAIN IS ESSENTIAL FOR THE ALIX DEVELOPMENTAL FUNCTION. IN ADDITION, WE SHOW THAT ALIX IS ASSOCIATED WITH THE ENDOSOMAL PATHWAY IN COLLABORATION WITH THE ESCRT MACHINERY INVOLVED IN THE SORTING OF THE MEMBRANE PROTEINS AND THE BIOGENESIS OF THE MULTIVESICULAR BODY (MVB). THE DISRUPTION OF THE COMPONENTS OF THE MVB SORTING MACHINERY LEADS TO DIFFERENT PHENOTYPES SUGGESTING EITHER ADDITIONAL ROLES IN OTHER PROCESSES FOR SOME COMPONENTS OR A DIFFERENTIAL IMPLICATION OF THE PROTEINS IN THIS PROCESS.
