Dynamique de localisation de la kinase mitotique Aurora-A et caractérisation de la protéine passagère TD-60 au cours de la mitose.

Abstract : Many kinases control cell cycle progression. In eukaryotic organisms, Aurora-A and Aurora-B kinases have highly similar structure and play fundamental roles during mitosis. Aurora-A kinase localizes on centrosomes and is involved in centrosome cycle and mitotic spindle formation. Aurora-B kinase belongs to the chromosomal passenger protein family : it localizes on centromeres and move to the spindle midzone in anaphase and concentrates on the contractile ring in cytokinesis. Aurora-B is responsible for the mitotic phosphorylation of histone H3 Serine 10 residue. By using a pseudo-genetic approach in human cells, we target, inside the N-terminal region of Aurora-A, the domain involved in the centrosomic localization of Aurora-A. These experiments also demonstrate that both catalytic domains of Aurora-A and Aurora-B contain a signal of centromeric localization. However, in opposite to Aurora-B, the catalytic domain of Aurora-A is not able to transfert from centromeres to spindle midzone in anaphase. Moreover, these results show that Aurora-A partly restores the mitotic functions of Aurora-B. In addition, we have identified and cloned the gene coding for the passenger protein xTD-60 of Xenopus laevis. We have expressed several truncated forms of xTD-60, in order to produce specific antibodies against xTD-60. Co-sedimentation experiments of xenopus eggs mitotic extracts proteins and cellular immunolocalization experiments provide new informations allowing to consider for xTD-60 other functions than those associated to passenger proteins.
