Etudes liées à la vitrification sans fracture de solutions cryoprotectrices

Abstract : Cryopreservation by vitrification could provide a general solution to the problem of long term organs cryoconservation. This method requires the use of non toxic, though high concentrations of cryoprotective agents in combination with rapid cooling rates, in order to prevent ice crystallization into tissues. We used a differential scanning calorimeter to determine the critical cooling rates necessary to vitrify the cryoprotective agents. A study undertaken on ethylene glycol informed us more precisely of the concentration from which aqueous solutions of this compound have an amorphous state sufficiently stable to be obtained experimentally. The study of the vitrification method has shown that annealing avoids the appearance of fractures during vitrification in large cryoprotective solution volumes. Work made in the case of VM3 (21st Century Medicine) is detailed. Obviously, the glass stability and the quantity of ice crystallized on warming are under the influence of the annealing temperature and duration. A large analysis on the influence of the container type and shape on the glass stability was started in parallel, but the limits of our cryogenic device pushed us to develop a new automated cryostat, allowing to make systematic studies under reproducible conditions. This cryostat was entirely conceived and built in the CRTBT. It represents the prototype of what we wish to propose for clinical studies of biological systems using cryopreservation. Its accuracy and ease of use allowed us to highlight various events taking place during the vitrification and the reheating of our samples.
