Thermodynamique et cinétique dans les macromolécules : apports de la microcalorimétrie AC de très haute résolution

Abstract : The aim of this work is to outline the possibilities offered by AC microcalorimetry for studies on thermodynamics and kinetics in macromolecules. As a first part a simple system (A+B) is described using both thermodynamic and kinetic approaches. Then we describe the main calorimetric techniques. As a third part we describe the experimental setup designed for this study. Based on AC calorimetry method, it requires as few liquid as 5 µl. It allows 1 ppm resolution on heat capacity measurement, and its frequency range covers two decades (0,05 – 5 Hz). After computer simulations on a simple system , we present several experimental results. Solid-liquid transition of an organic liquid exhibits irreversibility effects. Thanks to the wide frequency range available with our AC microcalorimeter. Spécifique heat spectroscopy experiments are lead on a polymer sample, and kinetics effects can be studied ; Heat-induced denaturation is observed on 25 µg of a globular protein, showing good perspectives for a frequency study.
