Organisation structurale et fonctionnelle des chromosomes

Abstract : A combination of two novel micropipette based techniques were used to measure the elasticity of an individual mitotic chromosome assembled in Xenopus egg extract. Our data show that the mitotic chromosome is not a homogenous object. It is constituted of two distinct parts: a soft chromatin envelope and inner rigid cores. The SMC proteins play a key role in the organization of the cores. We have also shown that both DNA and the proteins, but not RNA, are necessary for the maintenance of the chromosome structure. The energy landscape of the mitotic chromosome was also studied and new information for these complex objects was obtained. A dynamic model for the mitotic chromosome assembly was proposed. In this model the SMC proteins are responsible for the rod-like shape of the mitotic chromosomes while the bivalent ions Ca++ and Mg++ are essential for chromosome condensation.
