La médecine conduite par biomarqueurs pour le développement des nouveaux anti-cancéreux

Abstract : In 2015, cancer accounted for 8.8 million deaths worldwide, including 150,000 in France. Cancer cells have several characteristics, each of them might be the target of biomedical drugs that represent a promising approach for the treatment of such diseases. Despite having marketing authorizations, many targeted therapies are to date disappointing. The average cost for this kind of therapeutics in France is estimated at €50,000 yearly per patient. Thus, their ineffectiveness represents a considerable financial loss. The development of new therapeutic strategies is therefore necessary in order to reduce their societal impact. However, many of them fail at the clinical development stage. The optimization of preclinical study models and, above all, the integration of genomic medicine into clinical trials are promising ways to improve the chances of success of new drug candidates. Genomic medicine, in fact, makes it possible to look at a patient's genome in order to determine the pharmacological and pharmacokinetic response to a given treatment, thus giving the right treatment, at the right dose, to the right patient. However, the ethical aspect should not be neglected given the large amount of information obtained by high throughput sequencing of the human genome.
