ANALYSE STRUCTURALE DE PROTEINES DE L'ENVELOPPE NUCLEAIRE IMPLIQUEES DANS DES PATHOLOGIES GENETIQUES

Résumé

Inner nuclear membrane proteins and intermediate filaments that bind to chromosomes are proposed to play an important role in nuclear assembly. They might also mediate the nuclear positioning of individual chromosomes, and thus influence gene expression. Recently, it has been shown that these proteins are mutated in several pathologies. We aim at determining the 3D structure of nuclear envelope proteins in complex with their biological partners in order to shed light on the molecular mechanisms of the linked pathologies. During my PhD, I focus my work on certain proteins of the nuclear envelope: lamin A, emerin and MAN1. In order to understand the physical basis of MAN1-R-Smads interaction, I characterized the C-terminal nucleoplasmic region of MAN1, which is responsible for Smad binding. This region exhibits an N-terminal globular domain adopting a WH fold, which binds to DNA. Modeling the complex between DNA and the winged helix domain of MAN1 shows that the regions involved in DNA binding are essentially distinct from those reported to be involved in Smad binding. This suggests that MAN1 binds simultaneously to R-Smads and their targeted DNA sequences. In a second project, I tried to characterize the direct interactions between the C-terminal domain of A-type lamins and two partners, emerin and SREBP1. We don't corroborate there interactions with biochemical assays. During my PhD, I worked also on a novel mutation, which affects the C-terminal globular domain of lamins A and C, R439C. This mutation presents a distinctive form of FPLD with progeroid syndrome. At a molecular level, the three-dimensional structure and thermal stability of the C-terminal domains of mutant and wild-type lamins A/C are similar. However, the introduction of an additional cysteine provokes the formation of disulfide-mediated lamin A/C oligomers in vitro. This oligomerisation affects interaction properties of C-terminal domain with DNA. In conclusion, this novel mutation could suggest a novel pathogenetic mechanism for laminopathies.
Les protéines de la membrane nucléaire interne ainsi que les lamines, filaments intermédiaires nucléaires, jouent un rôle majeur dans l'assemblage et l'architecture du noyau. Ils interviennent aussi probablement dans le positionnement des chromosomes et influencent ainsi l'expression des gènes. Récemment, il a été montré que ces protéines sont mutées dans plusieurs pathologies. Notre objectif est de déterminer la structure tridimensionnelle des protéines de l'enveloppe nucléaire en complexe avec leurs partenaires biologiques afin de proposer des mécanismes moléculaires pour les pathologies correspondantes. Pendant ma thèse je me suis focalisée sur certaines protéines de l'enveloppe nucléaire : les lamines de type A, l'émerine et MAN1. Afin de comprendre les caractéristiques de l'interaction MAN1-R-Smads, j'ai caractérisé la région C-terminale de MAN1, responsable de la liaison. Cette région est composée d'un domaine N-terminal qui adopte un repliement structural de type WH qui lie l'ADN. La modélisation du complexe WH-ADN montre que les régions impliquées dans la liaison à l'ADN sont distinctes de celles prédites pour l'interaction avec les R-Smads. MAN1 peut donc lier simultanément les R-Smads et l'ADN. En parallèle, j'ai essayé de caractériser des interactions directes entre la partie C-terminale de la lamine de type A et l'émerine ou SREBP1. Nous n'avons pu confirmer ces interactions. J'ai aussi travaillé sur une nouvelle mutation de l'Ig fold de la lamine A, R439C, à l'origine d'une FPLD avec des symptômes progéroides. A un niveau moléculaire, la structure tridimensionnelle et la stabilité thermique de l'Ig fold sont similaires. Par contre, l'introduction d'une cystéine provoque la formation d'oligomères de l'Ig fold par les ponts disulfures qui affecte les propriétés d'interaction avec l'ADN. En conclusion, cette mutation pourrait être à l'origine d'un nouveau mécanisme pathologique des laminopathies.
